:
Role of (-)Epicatechin modulation of hepatic detoxifying enzymes against CCl4 induced liver damage in rats (-)Epicatechin (EP) is a member of polyphenols in the flavonoid family used as an antioxidant. EP may reduce the risk of oxidative stress induced by hepatotoxicity that is effects on the metabolites and detoxification. The aim of this study was designed to evaluate the protective effect of EP on liver fibrosis or cirrhosis induced by CCl4 in rats. 50 male rats were randomly divided into 5 groups (10 rats/group). The groups were treated as follows: Group I served as normal control; Group II (EP group) were administrated EP by orally 20 mg/kg b.w. for 3 weeks; Group III (CCl4 group) were injected i.p. of 1 ml/kg b.w. CCl4 2/3 weeks; Group IV (CCl4&EP) were injected the same doses of CCl4 and EP; Group V (CCl4&Sily) were injected the same dose of CCl4 2/3 weeks and 50 mg/kg b.w. SILY by orally for 3 weeks. Liver functions, oxidative stress biomarkers, detoxified enzyme biomarkers, anti-inflammatory biomarker and histopathological changes were evaluated. AST, ALT, ALP activities and MDA level were reduced, where serum albumin level was elevated in rats induced hepatotoxicity and treated with EP compared with rats injected i.p. CCl4. EP administration as treated of hepatotoxicity caused reduction in MDA while GSH, CAT, and GST were increased as compared with normal rats. NOS and CYP 450 as biomarker of detoxified enzyme were lower in rats induced liver damage and treated with EP or SILY. Histological examination revealed that inflammatory infiltrates was observed in liver tissues of rats received CCl4 that ameliorative when rats administrated EP or SILY. The rats treated with EP or SILY ameliorated the hepatotoxicity by inhibiting oxidative stress and increasing GST, NOS and CYP 450. EP as SILY could be administrated as the treated to decrease liver injury
